Retatrutide: Promising New Research in Treating Obesity

What Is Retatrutide?
Eli Lilly, which currently markets diabetes and weight-loss medications, is conducting clinical trials of retatrutide, a potential next-generation obesity and diabetes drug. Because it’s still in clinical trials, it hasn’t yet received approval from the U.S. Food and Drug Administration (FDA).
Retatrutide is what Eli Lilly calls a “triple G” drug, a once-weekly hormone receptor agonist that targets three receptors:
- Glucagon-like peptide-1 (GLP-1): This receptor slows digestion and controls appetite. This process helps you feel fuller longer, which means you consume fewer calories.
- Gastric inhibitory polypeptide (GIP): The hormone GIP helps lower your blood sugar and improve fat metabolism after meals. Improved fat processing and storage can make weight loss easier.
- Glucagon (GCG): Glucagon helps your body burn stored fat, enabling you to burn more calories, reduce your overall body fat, and improve your metabolism.
Because retatrutide targets these three hormone receptors, it’s considered a triple agonist. This important characteristic sets Retatrutide apart from drugs currently on the market, which target one or two hormone receptors. In contrast, honing in on all three receptors has a more powerful effect, promoting appetite reduction, blood sugar control, energy balance, metabolism, and fat burning.
Promising Results from Clinical Trials
Retatrutide can be a game-changer in treating obesity, type 2 diabetes, and fatty liver disease. Here are some of the latest findings from clinical trials assessing the safety and efficacy of Retatrutide:
Weight Loss and Obesity Management
Retatrutide may be a viable alternative for people who have had difficulty losing weight with other medications or methods. One study showed that retatrutide helped people taking the highest doses lose more than 20% of their body weight after 11 months. Even at lower doses, weight reductions of five percent or more occurred.
Participants also showed improved metabolism and heart health. In terms of safety, the study found that retatrutide side effects and tolerability were comparable to other GLP-1 and GIP therapies currently available for weight loss and type 2 diabetes.
Another study enrolled more than 400 individuals with obesity and knee osteoarthritis. After 16 months of treatment with retatrutide, participants’ average weight loss was 71 pounds, or 29% of their body weight. The researchers also observed a 76% reduction in knee pain over the same period. One implication of these findings is that retatrutide may enhance mobility among people who attempt to exercise or increase their physical activity to lose weight.
Type 2 Diabetes
Research suggests that Retatrutide can lower blood sugar associated with type 2 diabetes and promote long-term weight loss. One clinical trial evaluated the drug’s safety and effectiveness in lowering blood sugar and improving body weight.
At varying doses, retatrutide led to significant improvements in blood sugar levels and body weight. More than 80% of participants achieved an A1C below 6.5%, which is in the prediabetes range. Thirty-one percent of participants had A1C levels drop below 5.7%, which is considered healthy for most adults.
Regarding weight loss, an overall 17% reduction was observed across doses at week 36 of the study, and participants did not experience a weight-loss plateau. The findings suggest that retatrutide’s control of blood sugar is significantly correlated with its effect on body weight. This association is stronger than that observed with other medications tested for both type 2 diabetes and body weight.
Non-Alcoholic Fatty Liver Disease
Other researchers are investigating the use of Retatrutide in treating non-alcoholic fatty liver disease (NAFLD). The drug could slow or stop the progression of NAFLD, a complication in some people with excess weight.
One study examined retatrutide’s effectiveness at treating individuals with obesity and NAFLD. They found that the medication significantly reduced liver fat content at weeks 24 and 48 of the trial. Fat content decreased by 70-80%, depending on the dosage.
At week 48, more than 85% of the study participants experienced a reduction or reversal of liver fat accumulation. At the highest doses, that rate reached 90%.
Is Retatrutide Available Now?
Retatrutide is still in late-phase clinical trials and has not yet received FDA approval. Therefore, it’s not yet available on the market. While experts can’t predict when the drug will become available, it’s unlikely to happen until late 2026, or even 2027 or 2028. When the drug does hit the market, it’s intended to be taken weekly.
